Not sure if dlc character portraits on otherwise customizable avatars should count. If so, the Trauma Center guy has a Ryu number via the HD re-release of Etrian's Odyssey. The precedent might give Vox Machina a Ryu Number, but Pillars of Eternity is artfully self-contained.

Afaict, even omitting Rule 1 (videogames only), characters in the Glorantha setting don't have a Ryu Number. Cthulhu Wars has a spin-off called Glorantha: The Gods War, and Chaosium bucked the trend of adapting everything into a D&D supplement.
